    New Natural Diterpene-Type Abietane from Tetradenia riparia Essential Oil with Cytotoxic and Antioxidant Activities

    Tetradenia riparia (Hochstetter) Codd belongs to the Lamiaceae family and it was introduced in Brazil as an exotic ornamental plant. A previous study showed its antimicrobial, acaricidal and analgesic activities. Two compounds were isolated from essential oil of T. riparia leaves and identified as 9 beta, 13 beta-epoxy-7-abietene (1), a new one, and 6,7-dehydroroyleanone (2), already reported for another plant. The structure of these compounds was determined by spectroscopic analysis and by comparison with literature data. The cytotoxic activities of the essential oil and compounds 1 and 2 were determined by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, and by tumor cells MDA-MB-435 (human breast carcinoma), HCT-8 (human colon), SF-295 (human nervous system) and HL-60 (human promyelocytic leukemia). The essential oil and compound 1 showed high cytotoxic potential of the cell lines SF-295 (78.06% and 94.80%, respectively), HCT-8 (85.00% and 86.54%, respectively) and MDA-MB-435 (59.48% and 45.43%, respectively). Compound 2 had no cytotoxic activity. The antioxidant activity was determined by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), beta-carotene-linoleic acid system and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. The inhibitory concentration (IC50 in mu g mL(-1)) for essential oil and compound 2 was, respectively 15.63 and 0.01 for DPPH; 130.1 and 109.6 for beta-carotene-linoleic acid and 1524 and 1024 for ABTS. Compound 1 had no antioxidant activity. By fractioning the oil, it was possible to identify two unpublished compounds: 1 with high cytotoxic potential and 2 with high antioxidant potential

    The Ninth Data Release of the Sloan Digital Sky Survey: First Spectroscopic Data from the SDSS-III Baryon Oscillation Spectroscopic Survey

    The Sloan Digital Sky Survey III (SDSS-III) presents the first spectroscopic data from the Baryon Oscillation Spectroscopic Survey (BOSS). This ninth data release (DR9) of the SDSS project includes 535,995 new galaxy spectra (median z=0.52), 102,100 new quasar spectra (median z=2.32), and 90,897 new stellar spectra, along with the data presented in previous data releases. These spectra were obtained with the new BOSS spectrograph and were taken between 2009 December and 2011 July. In addition, the stellar parameters pipeline, which determines radial velocities, surface temperatures, surface gravities, and metallicities of stars, has been updated and refined with improvements in temperature estimates for stars with T_eff<5000 K and in metallicity estimates for stars with [Fe/H]>-0.5. DR9 includes new stellar parameters for all stars presented in DR8, including stars from SDSS-I and II, as well as those observed as part of the SDSS-III Sloan Extension for Galactic Understanding and Exploration-2 (SEGUE-2). The astrometry error introduced in the DR8 imaging catalogs has been corrected in the DR9 data products. The next data release for SDSS-III will be in Summer 2013, which will present the first data from the Apache Point Observatory Galactic Evolution Experiment (APOGEE) along with another year of data from BOSS, followed by the final SDSS-III data release in December 2014.Comment: 9 figures; 2 tables.     In vitro antifungal, and anti-elastase activity of some aliphatic aldehydes from Olea europaea L. fruit

    Olea europaea preparations are traditionally employed in a variety of troubles, including skin infections. Olive extracts and some of their pure compounds have shown antimicrobial activity in vitro. The present study deals with the antifungal activity of some aliphatic aldehydes from olive fruit [hexanal, nonanal, (E)-2-hexenal, (E)-2-heptenal, (E)-2-octenal, (E)-2-nonenal] against Tricophyton mentagrophytes (6 strains), Microsporum canis (1 strains) and Candida spp. (7 strains). The capability of these substances to inhibit elastase, a virulence factor essential for the dermatophytes colonization, and their cytotoxicity on cultures of reconstructed human epidermis, are also described. Aldehydes tested, inhibited the growth of T. mentagrophytes and M. canis in the range of concentration between < 1.9 and 125 mu g/ml; the unsaturated aldehydes showed the most broad spectrum of activity in that inhibited all strains tested. None of the aldehydes exhibited activity against Candida spp. strains. (E)-2-octenal and (E)-2-nonenal inhibited the elastase activity in a concentration-dependent manner; the anti-elastase activity suggests an additional target of the antimicrobial activity of these compounds. Aldehydes were devoid of cytotoxicity on cultures of human reconstructed epidermis. The antifungal activity of the aldehydes from olive fruit here reported, substantiates the use of olive and olive oil in skin diseases and suggests' that these natural compounds could be useful agents in the topical treatment of fungal cutaneous infections.     Reaction of corn genotypes to eyespot caused by Kabatiella zeae<br>Reação de genótipos de milho à mancha ocular causada por Kabatiella zeae

    The objective of this study was to evaluate the reaction of corn genotypes to eyespot caused by Kabatiella zeae under natural field conditions. The assay was carried out during the 2006/2007 growing season, in Pato Branco-Paraná State, consisting of 33 hybrids and two sowing seasons, on 10/13 and 12/27/2006, respectively; and in the 2007/2008 growing season, in the same place, with 10 hybrids and two sowing seasons, on 10/20 and 11/12/2007, respectively, and in Palma Sola-Santa Catarina State, with 8 hybrids and one sowing season, on 09/30/2007. Disease intensity was determined by means of severity evaluations. The observed corn plant reactions allowed genotypes with different levels of eyespot resistance to be discriminated. Hybrids AG 9020 and SPRINT were the most susceptible to the disease, while AS 1565 and DKB 234 were the most resistant; this behavior was repeated in both years and growing seasons. It was concluded that the variation in susceptibility to eyespot among the hybrids tested, may have been influenced by ambient conditions.<p><p>O objetivo deste trabalho foi avaliar a reação de genótipos de milho quanto à mancha ocular causada por Kabatiella zeae sob condições naturais de infecção. Os ensaios foram instalados na safra 2006/2007, no município de Pato Branco, PR, com 33 híbridos e com duas épocas de semeadura, dia 13/10 e 27/12/2006, respectivamente; e na safra 2007/2008, no mesmo município, com 10 híbridos e com duas épocas de semeadura, dia 20/10 e 12/11/2007, respectivamente e no município de Palma Sola, SC, com oito híbridos e com uma época de semeadura, dia 30/09/2007. A intensidade da doença foi determinada pela avaliação de severidade em todos os híbridos de milho, em diferentes estádios de desenvolvimento. As reações de milho observadas permitem separar estatisticamente genótipos com diferentes níveis de resistência à mancha ocular. Os híbridos AG 9020 e SPRINT apresentaram maior suscetibilidade à doença, e AS 1565 e DKB 234, maior resistência, repetindo este comportamento nos dois anos e épocas de cultivo. Concluiu-se que a variação na suscetibilidade à mancha ocular entre os híbridos de milho testados, pode ter sido influenciada pelas condições ambientais

    Mechanism of antibacterial action of three monoterpenes

    In the present paper, we report the antimicrobial efficacy of three monoterpenes [linalyl acetate, ()menthol, and thymol] against the gram-positive bacterium Staphylococcus aureus and the gram-negative bacterium Escherichia coli. For a better understanding of their mechanisms of action, the capability of these three monoterpenes to damage biomembranes was evaluated by monitoring the release, following exposure to the compounds under study, of the water-soluble fluorescent marker carboxyfluorescein from unilamellar vesicles with different lipidic compositions (phosphatidylcholine, phosphatidylcholine/phosphatidylserine [9:1], phosphatidylcholine/ stearylamine [9:1], and phosphatidylglycerol/cardiolipin [9:1]). Furthermore, the interaction of the terpenes tested with dimyristoylphosphatidylcholine multilamellar vesicles as model membranes was monitored by means of differential scanning calorimetry. Finally, the results were related to the relative lipophilicity and water solubility of the compounds examined. Taken together, our findings lead us to speculate that the antimicrobial effect of ()menthol, thymol, and linalyl acetate may result, at least partially, from a perturbation of the lipid fraction of microorganism plasma membrane, resulting in alterations of membrane permeability and in leakage of intracellular materials. Besides being related to physicochemical characteristics of the drugs (such as lipophilicity and water solubility), this effect seems to be dependent on lipid composition and net surface charge of microbial membranes. Furthermore, the drugs might cross the cell membranes, penetrating into the interior of the cell and interacting with intracellular sites critical for antibacterial activity
